TY - THES AB - In the frame of this thesis, a concept generally applicable to functional proteomics has been developed which opens up new possibilities for proteome analysis by using reversibly binding inhibitors for affinity-based tagging of protein families. Since a covalent bond between protein and reporter group is necessary, the reversible inhibitor must be equipped with an additional reactive group, e.g., a photo-reactive group. Isoquinolinesulfonamides, which competitively inhibit a broad range of kinases by occupying the ATP-binding site, were chosen to prove the applicability of this concept.
